    Abstract The reduction of landfill gas emissions is a central issue of the Directive 99/31/EC. Biofilters and biocovers have been identified as an alternative and cost-effective technologies to mitigate impacts due to CH4 and NMVOCs emissions. The Life Cycle Assessment demonstrates the environmental sustainability of biofiltration systems, with the aim of improving the environmental impact indicators such as Global Warming (-10.75% for Biofilter and -11.60% for Biocover) and Photochemical oxidation (-7.97% for Biofilter and -8.61%. for Biocover). This paper shows that these treatment technologies are effective for methane oxidation when the calorific value of the LFG is low, thus they maximize the amount of treated gas during the after-care phase

    Competencies based innovative learning solutions for co-development of climate services in West Africa

    Abstract. In developing countries, and particularly in West Africa, the role of Climate Services (CS) for sustainable development is growing thanks to wide spreading collaboration among European institutions, including National Meteorological and Hydrological Services (NMHS) research centers, universities, and homologue local institutions. Operationally, the implementation of CSs in developing countries is mainly pivoted on NMHS, which, according to the World Meteorological Organization (WMO), are dramatically affected by unmet learning demand. The global scale of learning needs for co-development of CSs calls for innovative solutions and a range of flexible modalities to reach learners in a variety of ways, and for sharing resources and successful strategies within the global education and training community. In order to harmonize expected learning outcomes, WMO defined a competency framework (CF) for CSs to be used in the implementation of training initiatives and knowledge sharing tools. This paper presents the strategic and methodological approach adopted in the implementation of the TOPaCS, a new knowledge-based distance learning initiative, aiming to provide a flexible learning environment within the CSs CF of WMO ensuring coherence with other WMO education initiatives (Global Campus, other RTCs, etc.). The methodological approach adopted is based on the competency-based approach to training, where competencies are composed by elements of knowledge and skill. TOPaCS integrates the WMO CF for CSs into a taxonomy co-designed with stakeholders at different levels, and allows the definition of learning paths, which are a further interactive opportunity for co-development of CSs within the TOPaCS learning ecosystem. Indeed, the approach aims also to guide further instructional strategies and assessments and becomes a starting point to build a common language enabling a better cooperation and exchange between the different CSs training initiatives

    [IT] In questo contributo si focalizza l’attenzione sulla ricostruzione grafica e virtuale del patrimonio e sulle sensazioni che la ricostruzione evoca nel fruitore del messaggio culturale. La ricostruzione si attua mediante la collaborazione tra archeologo e illustratore e costituisce un momento di verifica visiva dell’interpretazione archeologica e uno strumento di comunicazione del dato archeologico fruibile a più livelli. Ciò avviene grazie allo scambio costante di informazioni (dati scientifici e proposte di ricostruzione) e il confronto che ne deriva determina i metodi e le fasi di avanzamento del progetto. Il metodo si basa sull’integrazione delle immagini 3D con il disegno tradizionale ed è finalizzato alla trasposizione del dato archeologico, in modo tale da garantire alla ricostruzione il duplice requisito di soddisfazione estetica e credibilità scientifica.[EN] This paper focuses on the graphic reconstruction process and on the emotional response that the reconstructions stimulate. The reconstruction process is a collaboration carried out by archaeologist and illustrator and shall be considered as an instrument for both the visual verification of the archaeological interpretation and the spread out of the archaeological data to different kinds of beneficiaries. This process is made possible by the continuous exchange of information (scientific data and reconstruction hypotheses) and the debate originating from this helps in setting methods and schedules of the reconstruction process.     Untangling the extracellular matrix of idiopathic epiretinal membrane: a path winding among structure, interactomics and translational medicine

    Idiopathic epiretinal membranes (iERMs) are fibrocellular sheets of tissue that develop at the vit-reoretinal interface. iERMs consist of cells and extracellular matrix (ECM) formed by a complex array of structural proteins and a large number of proteins that regulate cell-matrix interaction, matrix deposition and remodelling. Many components of the ECM tend to produce a layered pat-tern that can influence the tractional properties of the membranes. We applied a bioinformatics approach on a list of proteins previously identified with an MS-based proteomic analysis on sam-ples of iERM to report the interactome of some key proteins. The performed pathway analysis highlights interactions occurring among ECM molecules, their cell receptors, and intra or extra-cellular proteins that may play a role in matrix biology, in this special context. In particular, integ-rin β1, cathepsin B, epidermal growth factor receptor, protein-glutamine gam-ma-glutamyltransferase 2, and prolow-density lipoprotein receptor-related protein 1 are key hubs in the outlined protein-protein cross-talks. A section on the biomarkers that can be found in the vitreous humor of patients affected by iERM and that can modulate matrix deposition is also pre-sented.     Induced sputum is a reproducible method to assess airway inflammation in asthma.

    To evaluate the reproducibility of induced sputum analysis, and to estimate the sample size required to obtained reliable results, sputum was induced by hypertonic saline inhalation in 29 asthmatic subjects on two different days. The whole sample method was used for analysis, and inflammatory cells were counted on cytospin slides. Reproducibility, expressed by intra-class correlation coefficients, was good for macrophages (+0.80), neutrophils (+0.85), and eosinophils (+0.87), but not for lymphocytes (+0.15). Detectable differences were 5.5% for macrophages, 0.6% for lymphocytes, 5.2% for neutrophils, and 3.0% for eosinophils. We conclude that analysis of induced sputum is a reproducible method to study airway inflammation in asthma. Sample sizes greater than ours give little improvement in the detectable difference of eosinophil percentages

    The authors revise the concept of anaplastic large cell lymphoma (ALCL) in the light of the recently updated WHO classification of Tumors of Hematopoietic and Lymphoid Tissues both on biological and clinical grounds. The main histological findings are illustrated with special reference to the cytological spectrum that is indeed characteristic of the tumor. The phenotype is reported in detail: the expression of the ALK protein as well as the chromosomal abnormalities is discussed with their potential pathogenetic implications. The clinical features of ALCL are presented by underlining the difference in terms of response to therapy and survival between the ALK-positive and ALK-negative forms. Finally, the biological rationale for potential innovative targeted therapies is presented

    484. Preclinical Proof of Concept of Transcriptional Silencing and Replacement Strategy for Treatment of Retinitis Pigmentosa Due To RHODOPSIN Mutations

    Silencing and replacement strategy is a promising approach to overcome mutational heterogeneity of genetic defects. In autosomal dominant retinitis pigmentosa (adRP) due to rhodopsin gene (RHO) approximately 200 different mutations have been described, posing a challenge for the design of effective therapeutics.We designed a silencing and replacement strategy based on transcriptional silencing through an artificial zinc finger DNA-binding protein lacking effector domains (ZF6DBD), and tested both efficacy and safety in two animal models.In a murine model of adRP, we show that AAV-mediate retinal delivery (AAV2/8-CMV-ZF6-DBD) is associated with selective transcriptional silencing of the human mutated allele resulting in morphological and functional (Electroretinography, ERG a-wave and b-wave responses) rescue. We then tested the effect of transcriptional silencing in the porcine large pre-clinical model. Delivery of a low dose (AAV2/8-CMV-ZF6-DBD, 1×10e10 vector genomes, vg) of the ZF6 transcriptional silencer to the porcine retina resulted in robust transcriptional silencing of the endogenous porcine RHO transcript. Cell sorting of transduced photoreceptors showed an almost complete RHO transcriptional silencing effect (90% RHO transcriptional repression), underscoring the potency of the system. To determine the safety of the zinc-finger silencer we performed extensive RNA-seq analysis on treated and control retinae. The data sets generated demonstrate selective RHO gene transcriptional repression and a remarkably low number of differential expressed genes (DEGs), supporting specificity and thus, safety. The co-administration to the porcine retina of the AAV-ZF6 silencer (AAV2/8-CMV-ZF6-DBD) and the AAV-RHO replacement (5×10e11 vg, AAV2/8-GNAT1-HumanRHO) constructs resulted in a balanced silencing and replacement effect. This data support the use of zinc-finger based RHO transcriptional silencing for the development of a clinical trial for adRP patients

    Transcription factors (TFs) function by the combined activity of their DNA-binding domains (DBDs) and effector domains (EDs). Here we show that in vivo delivery of an engineered DNA-binding protein uncoupled from the repressor domain entails complete and gene-specific transcriptional silencing. To silence RHODOPSIN (RHO) gain-of-function mutations, we engineered a synthetic DNA-binding protein lacking canonical repressor domains and targeted to the regulatory region of the RHO gene. AAV-mediate retinal delivery at a low dose (AAV2/8-CMV-ZF6-DBD, 1×10e10 vector genomes, vg) in the porcine retina resulted in selective transcriptional silencing of RHO expression. The rod photoreceptors (the RHO expressing cells) transduced cells when isolated by FACS-sorting showed the remarkable 90% RHO transcriptional repression. To evaluate genome-wide transcriptional specificity, we analyzed the porcine retina transcriptome by RNA sequencing (RNA-Seq). The differentially expressed genes (DEGs) analysis showed that only 19 genes were perturbed. In this study, we describe a system based on a synthetic DNA binding protein enabling targeted transcriptional silencing of the RHO gene by in vivo gene transfer. The high rate of transcriptional silencing occurring in transduced cells supports applications of this regulatory genomic interference with a synthetic trans-acting factor for diseases requiring gene silencing in a large number of affected cells, including for instance a number of neurodegeneration disorders. The result support a novel mode of gene targeted silencing with a DNA-binding protein lacking intrinsic activity

    Antimicrobial resistance, biofilm synthesis and virulence genes in Salmonella isolated from pigs bred on intensive farms

    Salmonella is the second cause of foodborne infection in humans in the USA and Europe. Pigs represent the second most important reservoir for the pathogen and the consumption of pork meat is a major risk factor for human salmonellosis. Here, we evaluated the virulence patterns of eleven Salmonella isolated from pigs (carcasses and faces) bred in intensive farms in the north of Italy. The two serotypes identified were S. Typhimurium and its monophasic variant 1,4,5,12:i:-. None of the isolates was an ESBL producer, as confirmed also by PCR. However, the presence of a multidrug resistant pattern was evident, with all the isolates being resistant to at least to five antimicrobial agents belonging to various classes. Moreover, six out of eleven isolates showed important resistance profiles, such as resistance against colistin and ciprofloxacin, with nine to twelve recorded resistances. The isolates were negative for the biofilm synthesis test, while four different virulotypes were characterized. All the isolates showed the presence of invA, hilA, stn, ssrA, sipC. One sample also harbored ssaR and spvC genes. One strain was positive for all the virulence genes tested and was resistant to 12 antimicrobial agents. The present study contributes new data to the surveillance program for antibiotic resistance. Furthermore, the presence of eleven highly virulent isolates poses concern for human health in relation to their diffusion in the environment
